There's no Biblical account of this being the first time that John and Jesus met, but they were cousins (both of their mothers were sisters) and I think thay might have grown up with each other. Remember that John leaped in his mother's womb (Luke 1:41) when Mary met Elizabeth (Mary's sister), so being filled with the Holy Spirit John knew that the baby in Mary's womb was the Christ.

If you look in the verses after 29, John says that he didn't recognize or know Jesus. Luke 1:80 states that John grew up in the wilderness until he appeared publicly before Israel. John 1:33 says that John wouldn't have known who Jesus was expect God told him that the man who John saw the the Spirit come down on was the one who will baptize with the Holy Spirit and finally in v.34, John says that he has seen and testify that the man the Spirit came down on was the Son of God. So according to the scriptures after v. 29 and the fact that Luke 1:80 states that John grew up in the wilderness, I believe that this was the first time John saw Jesus.
